The use of lipase inhibitor drugs to reduce the absorption of lipids has been widely accepted by researchers worldwide and has been reported in some cases. However, as there are no data available on the pharmacokinetics of these drugs, it is necessary to conduct further studies to confirm these findings.
In this prospective study, we used data from the pharmacokinetics of orlistat and all the other lipase inhibitors in the United States.
This prospective randomized clinical trial was conducted at the University of Pennsylvania (PA), Pennsylvania, USA, to investigate the pharmacokinetics of orlistat and orlistat and all the other lipase inhibitors in patients.
A total of 1206 patients were enrolled in the study. The patients were randomly assigned to receive orlistat (60 mg, 200 mg, or 500 mg) (n=360) or all the other lipase inhibitors (n=360) (n=360) (see ).
The pharmacokinetic parameters were determined by bio-analytical method as described previously (Prelude, 2011).
The pharmacokinetic parameters of all the lipase inhibitors were similar and dose-response was observed with the andlistat. The most significant differences in the mean area under the concentration-time curve (AUC) were observed for the andlistat (9.1 vs 6.6, respectively), and the area under the concentration-time curve to infinity (AUC0-t) were significantly more than for the lipase inhibitors (6.6 vs 3.9, respectively). The area under the concentration-time curve to infinity was not significantly different among the three groups.
This study demonstrated that orlistat and the other lipase inhibitors could be used safely and effectively to reduce the serum concentration of lipids and to enhance the pharmacokinetics of lipids in patients with the diagnosis of obesity. The results of this study provide further evidence for the use of these drugs and suggest the possibility of new strategies to reduce the absorption of lipids.
